Small Molecule
Get smoother filing and transfer pathway for drug and small molecule development.
Drug development can bring unexpected complexity.
The chemistry is established. The regulatory pathway is mapped. The precedent runs decades deep. But that familiarity can make evolving risks an even bigger surprise.
Guidance shifts on impurity limits. An API supplier’s data doesn’t hold up to scrutiny. A method that performed well in development won’t transfer to the receiving site. A dissolution profile drifts, and batches start failing.
Every one of those changes can turn a straightforward program into rework, lost lots, and months you can’t get back. But if you get a partner to help you lock down the fundamentals early, you can be ready for anything.

CMC Strategy and Regulatory Submissions
Your CMC package is the argument for your product’s quality. We define the strategy early, then generate and organize the data that supports it—including the adaptation work that ex-US programs need before they’re ready for US submission.
- CMC and regulatory strategy for ANDA, 505(b)(2), and NDA pathways
- Data review and section authoring for Module 3 and quality summaries
- Bioequivalence strategy and supporting documentation
- Adaptation of ex-US programs for US submission

- Alignment with FDA, EMA, and ICH expectations
API & Analytical
Small molecule risk concentrates in the chemistry. We build the control strategy so your specifications and methods stand up to review and continue to perform through transfer and commercial supply.
- Synthetic route assessment and impurity control strategy
- Genotoxic and elemental impurity evaluation
- Specification setting and justification
- Analytical method development, validation, and lifecycle management
- Stability strategy and program design
- Critical material and vendor qualification
Tech Transfer and Scale-Up
Moving from development scale to commercial volume introduces risk at every handoff. We assess your controls, procedures, and process maturity against global expectations, then build the roadmap and provide hands-on support to execute it.
- Gap analysis against global regulatory expectations
- Tech transfer planning, protocols, and batch records
- CDMO selection oversight and readiness activities
- Process validation strategy and execution support
- Commissioning, qualification, and validation readiness
- Method transfer and comparability planning
Small Molecule Manufacturing and Supply Recovery
When manufacturing performance slips, margin and market position go with it. We identify root cause quickly, design corrective actions that satisfy regulators, and return your program to stable supply.
- Root cause investigation and CAPA development
- Batch failure and out-of-specification investigations
- Process robustness assessment and improvement
- Predictive and in silico modeling for process performance
- Remediation planning and regulatory response
- Post-approval change strategy and justification
Case Studies
Build a drug development program that’s ready for anything.
You don’t have to solve API strategy, filing, and commercial supply alone. We’ve taken small molecule products from route selection through commercial launch before, and we know where programs get stuck. As a USP company, we bring a standards-setting perspective and expertise from FDA reviewers to every engagement. Connect with us and let’s create a path forward together.
Frequently Asked Questions
What does the small molecule drug development process look like?
It runs from selection and API development through formulation, analytical method development, scale-up, validation, and filing. Each stage produces decisions that constrain the next. The synthetic route determines your impurity profile. Your impurity profile determines your specifications. Your specifications determine what you can defend at review and what your manufacturing process has to deliver every batch. Programs that treat these as one connected strategy move faster than programs that treat them as sequential handoffs.
When should we bring in support for CMC strategy?
Earlier than most teams expect. Route, material sourcing, and analytical decisions made in early development determine what you can defend later. Bringing strategy in early costs far less than reworking a program after a filing stalls. If you’re already further along, a focused gap analysis will tell you where you stand and what has to close before your next milestone.
Do you work on ANDA and 505(b)(2) programs?
Yes. We support generic sponsors and 505(b)(2) applicants on CMC strategy, bioequivalence documentation, specification justification, and the data package that has to hold up under review. We also help sponsors understand which pathway their program actually fits before they commit to one.
We developed our product outside the US. What does it take to file here?
Ex-US programs often need meaningful adaptation before they’re ready for US submission—differences in specification expectations, impurity qualification, stability design, and data presentation are common. We assess your existing package against FDA expectations, identify what has to be generated or reworked, and build the submission strategy from there.
How do you approach impurity control?
We start with the synthetic route, because that’s where the impurity profile originates. From there we work through identification, qualification, and control—including genotoxic and elemental impurities—and build specifications you can justify with data rather than defend with argument. Impurity strategy is one of the most common sources of late-stage findings in small molecule programs, and one of the most preventable.
Our product is losing batches. Can you help us find the cause?
Yes. We investigate root cause, design corrective actions that satisfy regulators, and help you return to stable supply. In one case, a client losing 10 percent of batches annually to dissolution failures had root cause identified within six weeks and the product back on market after an 18-month absence.
How do you support tech transfer to a CDMO?
We manage the technical risk, not just the logistics. That includes gap analysis, risk assessments, protocol and batch record authoring, method transfer, comparability planning, and oversight of readiness activities at the receiving site. We’ve transferred programs across sites and organizations without losing regulatory alignment or schedule.
Can you help with API supplier qualification and oversight?
Yes. Supplier and data integrity issues surface at the worst possible times—during inspection, or after a filing is already under review. We support supplier qualification, technical oversight, audit readiness, and data integrity assessment across your external network.
What is in silico modeling, and where does it help in small molecule manufacturing?
In silico modeling uses computational simulation to predict how your process will behave under different conditions. It helps you define design spaces, identify critical process parameters, and predict performance—often replacing experiments that would consume months of lab time and material. It’s especially useful in process development, scale-up, and investigating manufacturing performance problems.
How do you handle post-approval changes?
A well-built control strategy anticipates change. We help you plan and justify post-approval changes, manage comparability, and implement process improvements without disrupting supply or triggering avoidable regulatory burden.
Do you work on small molecules only, or other modalities as well?
Both. Our teams cover small molecules, biologics and TIDES, cell and gene therapies, and combination products. We match the right expertise to your product rather than applying a single template.
